Indication of Probitor:Used in the treatment of a number of conditions associated with hyperacidity:- gastroesophageal reflux (symptomatic)- erosive oesophagitis (treatment and relapse prevention)- duodenal and gastric ulcer (treatment and relapse prevention- ulcers associated with H. pylori infection (as part of combination therapy for treatment and relapse prevention)- ulcers associated with anti-inflammatories (treatment and relapse prevention)- Zollinger-Ellison syndrome
Action of Probitor:Probitor works by inhibiting certain cells involved in acid production to reduce the amount of acid produced in the stomach. This reduces erosion of the stomach cells and helps with the pain associated with peptic ulcer disease. It also helps in diseases where too much stomach acid is regurgitated back into the oesophagus.
Dose advice of Probitor:Probitor capsules must be swallowed whole with water, not chewed or crushed. The following are the recommended doses of Omeprazole for treating various diseases. It should be noted, however, that Probitor is only available in 20mg capsules that cannot be halved. In situations where smaller doses are required, another brand should be used.Symptomatic GORD- 10-20mg once daily - maximum 4 weeks treatment- symptom relief is usually rapid, if symptoms have not been controlled in four weeks, further investigation is requiredErosive oesophagitis- 20mg once daily- healing usually occurs in four weeks, but a further four weeks treatment may be necessary and is allowable- refractory oesophagitis may require once daily dosing of 40mg for eight weeks- after healing, maintenance therapy should be continued at 10mg once daily, being increased to 20mg once daily if necessaryH. pylori associated peptic ulcer disease- 40mg once daily or 20mg twice daily with appropriate antibiotic regime:- amoxycillin 500mg + metronidazole 400mg three times daily for two weeks; or- amoxycillin 1g + clarithromycin 500mg twice daily for one week; or- clarithromycin 250mg + metronidazole 400mg twice daily for one week- return of H. pylori infection is an indication for re-treatmentDuodenal ulcer- 20mg once daily for four to eight weeks- refractory duodenal ulcer may require once daily dosing of 40mg for eight weeks- after healing, maintenance therapy should be continued at 10-20mg once dailyGastric ulcer- 20mg once daily for four to eight weeks- refractory gastric ulcer may require once daily dosing of 40mg for eight weeks- maintenance therapy should be continued at 20mg once dailyNSAID associated ulcer- 20-40mg once daily with or without continuing NSAID treatment- healing usually occurs in four weeks, but a further four weeks treatment may be necessary and is allowable- maintenance therapy should be continued at 20mg once dailyZollinger-Ellison syndrome- initial dose is 60mg once daily- dose should be adjusted to individual requirements and continued as long as clinically necessary- when daily dose exceeds 80mg, dose should be divided

Common side effects of Probitor:Probitor is generally well tolerated, but the following adverse effects have been reported in less than 10% of patients:- diarrhoea- constipation- abdominal pain- nausea/vomiting- flatulence- headache
Uncommon side effects of Probitor:The following effects are seen in less than 1% of patients:- dizziness- oversleeping- insomnia- vertigo- rash - itching skin- malaise
